The 2.2 MDa molecular weight of the bacterial 70S ribosome, combined with its limited solubility (≤10 µM), present significant technical challenges to performing high-resolution studies of co-translational folding in ribosome-nascent chain complexes. We are therefore exploring and developing a range of strategies to enhance the sensitivity and resolution of these investigations, from varied isotopic labelling schemes to novel NMR pulse programs, sampling schemes and analysis methods.
