BLAST

We did a BLAST analysis using ncbi protein blast database of all four organisms including the human isoform a as a reference and obtained the below results. The results highlighted in red are the isoforms and species that were previously identified in the FASTA section.

Every homologue got a score depending on how close it is to the reference. The higher the score the more similar it is. E value shows the probability that the score is due to chance; where the lower the E value the less like likely similarity is due to chance.

The coverage shows the length of the sequence that could be aligned compared to the reference.

We have also included a phylogenetic tree that shows the possible evolutionary tree of LAT homologues and how closely they are related.

Both the graphical representation of the BLAST search and the phylogenetic tree show that the Homo sapiens isoform a sequence is closely related to rodent and primate sequences, but not so closely related to carnivores. The search showed that there is a high degree of similarity between each of the sequences highlighted in red that is not due to chance. This is proven by the low E values. The close relation between different organisms indicates evolutionary conservation and means the amino acids interact with similar molecules. Hence, this means there could be a common signalling pathway and function of LAT across many organisms.
